SeSaM (Sequence Saturation Mutagenesis) is a patented method to create diversity on the gene level higher than the diversity generated by conventional methods such as error-prone PCR. The fact is that random mutagenesis methods produce only one nucleotide exchange per codon which leads to 39.2% of possible mutations. If two nucleotide exchanges per codon are achieved, this diversity increases to above 90%. This is what SeSaM is based on.
